How can I ensure only one user at a time can open a file in an i-bay?

You shouldn't need to. Which kind of file are you talking about ? For some types, you might need to disable the oplock (see https://wiki.contribs.org/DB_Variables_Configuration), but you shouldn't have to change anything else
